If your job duties include the need to have a valid driver's license or your alcohol use/abuse that cause your DUI could be found to be "substantially related" to your job duties, then it could be reasonable for your employer to request some information and documents regarding your arrest and conviction. You should contact an attorney and provide everything to the attorney for an adequate evaluation of your situation.

If you have experience with legal processes in criminal court then establish a time line and line up your witnesses, get all discovery from the state and prepare the defense. There is nothing simple about a trial and the technical skill necessary to present evidence requires some knowledge of the system. If you lose the case you will have a conviction for your lifetime, surcharges in the thousands of dollars, fines, court costs and jail time or probation.

Not unless one of the tests is off or they were taken at different times. Your body processes alcohol at a rate that is dependent upon things like your weight, you stomach contents, your metabolism, etc. Therefore, if the tests are taken at different times, they will show different results.
